Potential risk of tamoxifen: gut microbiota and inflammation in mice with breast cancer

Objective Tamoxifen is an effective anti-tumor medicine, but evidence has been provided on tamoxifen-related inflammation as well its impact gut microbiota. In this study, we aimed to investigate tamoxifen-induced microbiota and alteration. Methods We established a BC xenograft mouse model using the MCF-7 cell line. 16S rRNA gene sequencing was used qRT–PCR, western blotting, cytometric bead array were inflammation-related biomarkers. Various bioinformatic approaches analyze data. Results Significant differences in microbial composition, characteristic taxa, microbiome phenotype prediction observed between control, model, tamoxifen-treated mice. Furthermore, protein expression of IL-6 TLR5 up-regulated mice, while mRNA Tlr5 Il-6, group, down-regulated colon. The concentration IFN-γ, IL-6, IL12P70 serum Moreover, correlation-based clustering analysis demonstrated that inflammation-negatively correlated including Lachnospiraceae-UCG-006 Anaerotruncus , enriched inflammation-positively Prevotellaceae_UCG_001 Akkermansia group. Finally, colon histologic damage Conclusion treatment significantly altered increased breast cancer mice model. This may be related intestinal epithelial barrier up-regulation.
